The Effect of Coculture on Development of 2-Cell Stage Mouse Embryos.

Early transfer has led to asynchrony of the endometrium-trophectoderm interaction between the embryo and uterine environment. A culture system which could prolong culture time and increase embryonic cleavage rate and viability would improve the success rate. The purpose of this study was to evaluate the in vitro developmental promoting effect of coculture system on 2-cell stage mouse embryos. METHODS: In this experiment, we have cocultured mouse embryos with Vero cells and with amniocytes in m-KRB and Ham`s F-10. RESULTS: In m-KRB, The hatching rate of embryos cocultured with Vero cells (71%) was significantly increased compared with that of embryos cocultured with amniocytes (36%). In Ham`s F-10, the development rate of 2-cell stage mouse embryos showed no significant differences in three groups (control 89%; Vero cells 92%; amniocytes 95%). And also the cell number of expanded blastocyst in experimental groups was significantly greater as compared with that of the control group (P<0.01). CONCLUSION: Coculture system improve the quality of mouse embryos and might increase the implantation and pregnancy rate.
