Detection of Human Papillomavirus in Cervical Neoplasia using the In situ Polymerase Chain Reaction and Hybrid Capture System.

Abstract
OBJECTIVE
To determine the prevalence of HPV infection in chronic cervicitis, cervical intraepithelial neoplasia [CIN] I, II, III and invasive cervical cancer using the in situ PCR and Hybrid capture system. METHODS: We studied on 70 patients who had undergone conization for cervical lesion with chronic cervicitis, CIN, invasive cervical carcinoma at our hospital between August 1996 and March 1998. We compared the efficacy of Hybrid CaptureTM [Digene, USA] system and in situ PCR for HPV DNA detection according to disease entity. RESULTS: Using the Hybrid capture system, the detection rates of HPV showed 33.3% in chronic cervicitis, 35.8% in CIN I, 53.9% in CIN II, 60.0% in CIN III and 70.3% in invasive cervical cancer. On the other hand the detection rates of HPV by in situ PCR system showed 40.0% in chronic cervicitis, 42.8% in CIN I, 77.0% in CIN II, 86.7% in CIN III and 84.6% in invasive cervical cancer. CONCLUSION: This study demonstrated that the detection rate of HPV using the in situ PCR [65.7%] is more sensitive and accurate than Hybrid capture system [50.0%].[p<0.005].
